Mike Brown suffered a fracture and ligament damage to his right thumb in Saturday's first test with South Africa and will return to England later this week for further medical consultation.
England Head Coach Stuart Lancaster said: "Our best wishes to Mike. He did really well on Saturday and is very disappointed to be out of the tour but he must remember he has had a fantastic season and should be rightly proud of all that he has achieved."
Brown will be replaced in the squad by Bath's Nick Abendanon.
During week commencing 22nd April Harlequins will launch an innovative, new Membership Scheme which will replace the existing Season Ticket Holder Programme. Further details of the scheme will be announced in due course.
